The great therapeutic potential of retinoic acid and its natural and synthetic derivatives has evoked interest among dermatologists during the last 50 years. The recent discovery of different types, subtypes and isoforms of nuclear retinoic acid receptors not only helps to clarify the action of retinoids at the molecular level, but it may also provide clues to a rational design of synthetic receptor-selective retinoic acid agonists/antagonists with a desired therapeutic spectrum of action and reduced adverse effects. This study provides a concise review of the pharmacological, toxicological and clinical aspects of topical retinoids in dermatology.
